چکیده
Several laboratories have reported on the properties and characteristics of RNA polymerase reactions with synthetic polyribonucleotides as templates (l-3), with general agreement that the synthetic polyribonucleotides are efficient templates for the synthesis of complementary polyribonucleotides. The use of synthetic polyribonucleotides of different base composition and size is of particular value for certain studies of the mechanism of action of RNA polymerase. The present communication reports studies with synthetic oligonucleotides of known composition and size. Characteristics of the stimulation of reactions involving polyadenylic acid and polyuridylic acid by complementary oligonucleotides suggest that oligonucleotides may act as chain initiators for the RNA polymerase. A preliminary communication on the effect of complementary oligonucleotides on the RNA polymerase reaction has already been reported by us (4).

Template Specificity of Oligonucleotides for Ribonucleic Acid Polymerase.
Recently attempts have been made to d&m: chemically the templates used as substrates in bacterial ribonucleic acid polymerase systems and to analyze the RX.1 products of such systems (l-9). Enzymatically synthesized polydeosynucleotides (2, 3, 6, 8) and polyribonucleotides (5, 7, 9, 10) permitted studies of the mechanism involved in the polyribonucleotide synthesis. It has been shown that an RX...

Formation of homopolymers of ribonucleotides by polynucleotide phosphorylase is well known (1, 2). In this reaction, a polymerization of ribonucleoside diphosphates occurs with the elimination of inorganic orthophosphate. Reactions involving formation of polyadenylic acid from adenosine triphosphate with elimination of an inorganic pyrophosphate group have been described with enzymes from sever...

A ribonuclease which degrades RNA specifically in RNA. DNA hybrid structures has been purified X00-fold from Escherichia co2i B and ZOO-fold from E. coli strain DllO pot A1, endo I-, fhy-). For maximal activity, the enzyme requires MgZ+ and the presence of a sulfhydryl reagent. The enzyme is capable of digesting more than 95% of the RNA in RNA .DNA hybrids to acid-soluble products. The products...
متن کاملSynthesis and Expression of Modified bFGF Gene in Escherichia coli Cells
A new strategy for construction of synthetic gene encoding human basic fibroblast growth factor comprising DNA annealing-ligation and augmentation by polymerase chain reaction was introduced. The sequence of the gene and corresponding amino acid chain were modified in order to increase stability of the protein. First, 300 bp and 160 bp fragments of the gene were assembled from 18 oligonucleotid...
متن کاملMiR-34-a acts as a suppressor in neuroblastoma progression by targeting CD44.
OBJECTIVE To verify whether micro ribonucleic acid 34-a can exert its negative effects in human neuroblastoma cells. METHODS The study was conducted at The Affiliated Hospital of Qingdao University, Qingdao, Shandong, China during 15 months (from March 2015 to about June 2016). Quantitative reverse transcription polymerase chain reaction was used to find the differences of micro ribonucleic a...
